Effects of Freezing and Supercooling on the I131Metabolism of Thyroid Tissue

Popis: Guinea pig thyroid lobes were frozen at –3 to – 5 C for 15 min, thawed, and incubated at 37 C with I131 iodide. Compared with control glands, kept at 2 C before incubation, there was less incorporation into thyroglobulin and a lower ratio of I131 DIT/MIT. Supercooling to – 8 C or freezing in the presence of dimethyl sulfoxide resulted in a normal pattern of iodination. The iodide-concentrating ability of pre-frozen tissue was only slightly lower than that of controls. Histologic studies indicated accelerated autolysis of colloid during incubation of pre-frozen tissue. The findings suggest that damage to specific cell structures by ice crystals leads to the observed defects in iodination.
