Popis: | An antiserum produced in a sheep against 11-desoxycortisol was fractionated using an ion-exchange column of DEAE cellulose. Using stepwise elution at fixed pH and increasing ionic strengths, three fractions were obtained. These fractions were tested for titer, affinity, and specificity. All three parameters studied showed differences between the various fractions. Fraction II showed a pattern similar to that of the whole antiserum. It is suggested that for optimization of radioimmunoassay, it may be advantageous to purify antisera in order to screen for immunoglobulin fractions with the best characteristics. |
