Popis: | The paper deals with the development, optimization, and validation of solid-phase radioimmunoassay (RIA) using magnetic particles for estimation of luteinizing hormone (LH) in human serum or plasma. 125I-LH was prepared by chloramine-T oxidation method and purified by HPLC. The activation of magnetic particles using 1,1′-carbonyldiimidazole (CDI) and coupling of these solid-phase particles with rabbit anti-LH were carried out. The assay was optimized and validated. The intra- and inter-assay variations were satisfactory. The recovery and dilution tests indicated accurate calibration and appropriate matrix. The present technique agreed well with currently used commercial kit (Siemens). These magnetic particles retain their characteristics during storage for 1 year at 4°C. The technique proved to be sensitive, specific, precise, and accurate for routine laboratory use. |
