In this paper, PDMS (Polydimethylsiloxane) is used to fabricate a simple microfluidic biochip through microelectromechanical process technology. The detection system is fabricated using the laser excitation light principle to detect the electrical signal of the microchannel bio-nucleic acid hybrid. In the PDMS production, the micro-channel wafer mother film was fabricated by the yellow light lithography process using SU-8 GM1075 thick film photoresist, and then the mold was transferred by a casting technique to bond with the slide substrate to prepare an experimental biochip. The biochip is implanted into the wafer using DNA with a fluorescent probe, and the injected surfactant is used to observe the fluorescence change through the DNA sample, using two different laser wavelengths, a green laser wavelength of 532 nm and a red laser wavelength of 635 nm. PSoC, to detect samples and compare the sensitivity of two different wavelength detection systems. This experiment is for fluorescence detection. The two kinds of fluorescent probes, HEX and Cy5, are used for detection and comparison. The samples are identified by voltage signals.
