Preventive ability of liver fibrosis by Trapa taiwanensis Nakai fruit skin extracts in angiotensin converting enzyme II (Ace2) knockout mice

Popis: | 107 The liver is one of the most important metabolic organ of human. It may be induced liver disease by reactive oxygen species (ROS) accumulation or microbe infection. Recent studies showed that the hot water extract of Trapa taiwanensis Nakai (HWETT) had well anti-oxidation function in inflammation. As a folk medicine, the HWETT has been used to protect liver diseases. In previous studies, Ace2 knockout mice B6;129S5-Ace2tm1Lex/Mmcd had been established as a liver disease animal model to induce liver fibrosis. In this study, B6;129S5-Ace2tm1Lex/Mmcd was used to induce liver fibrosis by carbon tetrachloride (CCl4), and treated with HWETT, analyzed the effect of fibrosis inhibition. Extracts of HWETT were analyzed by 2,2-diphenyl-1-picrylhydrazyl (DPPH) and conformed to be a radical scavenger. As observed, the higher the HWETT concentrations, the higher the radical scavenging activity detected. HWETT also can protect liver cell line HepG2 from H2O2 damage in vitro. Compared with non-treated cell, HWETT-treated HepG2 cell enhanced the survival rate by H2O2 damage from 25% increased to 75%. In antioxidant enzyme, HWETT-treated upregulated RNA level of superoxidase dismutase (SOD) and catalase (CAT). It showed the anti-oxidant activation of HWETT was effective in HepG2 cell. In vivo, HWETT can protect mouse from CCl4-induced liver damage. After 8 weeks of CCl4 treatment (50% CCl4/olive oil by 2.0 mL/kg bw/twice/wk), HWETT-fed mice (125 mg/kg bw/day) compared with non-fed mice, the enzyme activities of serum aspartate transaminase (AST) and alanine transaminase (ALT) of mice was decreased. Histopathological examination represented less of cell death and extracellular matrix accumulation in HWETT-treated mouse liver. HWETT-treated was also decreased in glutathione disulfide (GSSG) accumulation, and maintain the activity of SOD and CAT enzyme in liver. Gene expressions of Tnf-α, Il-6, CCL-2, and CCL-5 were decreased in HWETT-treated mice detected by RT-PCR. The decrease of mRNA expression results confirmed that HWETT can protect mice liver from liver inflammation. As assay the protein concentration of α-SMA by Western blot, α-SMA was decreased in HWETT-treated mouse. It demonstrated that the generation of fibroblast was inhibited. In conclusion, Trapa taiwanensis Nakai fruit skin extract can protect the liver from chemical-induced liver inflammation and damage, and has potential to be a healthy food or farm animal feed additives to protect the liver. |
