Analysis of Superoxide Dismutase Expression from Pumpkin in Enhancing Chilling Stress in Arabidopsis Transgene

Excessive expression of endogenous SOD in Arabidopsis thaliana, knock out of endogenous SOD in Arabidopsis thaliana, and SOD from pumpkin were transferred into Arabidopsis thaliana and using PCR and Western blotting were used to confirm the performance. Using qRT-PCR is used to analysis gene expression under chilling stress. AtSOD gene expression of analysis shows that AtSOD are excessive expression and atsod are determined to decrease expression. CmSOD gene expression of analysis shows that CmSOD are determined to express a large number of CmSOD gene. Analysis of CBF2 gene know that AtSOD, atsod, and CmSOD all show the highest performance at 192 hours, and are 328-fold, 10-fold, and 76-fold higher than NT. Analysis of AtRD29A gene know that AtSOD show the highest performance at 72 hour and are 13.6-fold higher than NT, CmSOD have performance and are 2.6-fold NT at 192 hours. But atsod show the lowest performance at 72 hour and are 0.3-fold NT. Analysis of AtRD29B gene know that AtSOD are 33-fold higher than NT at 6 hours, atsod are 22-fold higher than NT at 6 hours, and CmSOD are 150-fold higher than NT at 6 hours. AtSOD4, CmSOD6 and atsod2 are selected for 1 μM ABA treatment. Analysis of CBF2 gene know that AtSOD4 are 4-fold higher than NT at 72 hours, atsod2 are 12-fold higher than NT at 6 hours and CmSOD6 are 2-fold higher than NT at 6 hours. Analysis of AtRD29A gene know that AtSOD4, CmSOD6 and atsod2 are 25, 9 and 3-fold higher than NT at 192 hours. Analysis of AtRD29B gene know that AtSOD4 are 70-fold higher than NT at 72 hours, atsod2 are 87-fold higher than NT at 6 hours and CmSOD6 are 6-fold higher than NT at 6 hours. The results of total chlorophyll content analysis show that AtSOD and CmSOD transgenic plants are 1.3 to 1.8-fold and 1.2 to 2.4-fold higher than NT under chilling stress. Observing the appearance and color of the leaf show that there is no difference in the appearance of AtSOD and CmSOD under chilling stress, and the color of the leaves is also greener than NT. NBT staining analysis show that the stained area and color of AtSOD leaves are less and lighter under chilling stress. However, the leaves of CmSOD are scarcely stained at the beginning, and are stained lighter and less under chilling stress.
