Effects of various Laminaria japonica-derived fucoidan on biological properties in anti-lung cancer cells

Fucoidan is a sulfated polysaccharide which extracted from brown algae, and presents immunomodulatory and anti-cancer functions. Our previously studies demonstrated that the anti-tumor effects of fucoidan from different resources are different. Specifically, we found that Lj-fucoidan from Laminaria japonica contained lots of sorbitol, which reduced anticancer efficacy of fucoidan. In this study, we performed the simple dialysis method to remove the sorbitol from Lj-fucoidan and examined the characteristics and anticancer functions of various dialyzed Lj-fucoidan. We found that dialysis effectively removed sorbitol from Lj-fucoidan and enhanced Lj-fucoidan-induced transforming growth factor receptor β (TGFRβ) and Slug degradation in lung cancer cells. Especially, Lj-fucoidan dialyzed for 4 days (called D4-fucoidan) presents higher biological activities. We further examined the D4-fucoidan on cell cycle markers, proteins (TGFR and slug) degradation effects, and the intracellular reactive oxygen species (ROS) changes. Functional assay revealed that TLR2/4 played the pivotal roles on anticancer functions of D4-fucoidan. In addition, we found that combination of D4-fucoidan and cisplatin, a chemotherapy medication, enhanced the anti-cancer effects. In summary, dialysis can effectively remove the sorbitol from Lj-fucoidan as well as enhance the effects on Lj-fucoidan-induced TGFR and slug degradation. Moreover, D4-fucoidan may be a potential agent for enhancing cytotoxic effects of cisplatin.
