Popis: | 106 Ophiocordyceps sinensis is one of the important herbs medicine in the Chinese world, And it has been confirmed by many experiments that it is rich in a variety of biological active ingredients and pharmacological effects. However, as a result of over-collection, the output has been declining year by year, resulting in high prices, so that counterfeit products are widely circulated in the market of Ophiocordyceps sinensis, In recent years, more groups and members have started the projects in cultivating artificial Ophiocordyceps sinensis, yet it is found the status of artificial mass production even is not well know in the market and it is believed there would be some bottleneck still. Therefore, our study intends to find out the better way to do the verification and optimization of the isolation and detection technology of wild Ophiocordyceps sinensis, we are hoping to provide more useful information for future mass production purpose. Four strains of Ophiocordyceps sinensis, Paecilomyces hepiali, Paecilomyces sinensis, and Pochonia suchlasporia were isolated successfully via our study. By using the common primer pair, the sequence containing 18S, ITS1, 5.8S, ITS2, and 28S can be checked and analyzed. The ITS identification and sequence alignment analysis finally confirmed the isolates we separated - Ophiocordyceps sinensis, Paecilomyces hepiali, Paecilomyces sinensis and Pochonia suchlasporia all are matching to the database. In our research results, the multiplex PCR method was compared, and also its molecular fingerprint database was established. |
