Triterpenoids are the major active ingredients contributing to antidiabetic, antibacterial and anticancer activities of wild bitter melon. Traditional sample preparation used ultrasonic extraction, and analysis by high-performance liquid chromatography with evaporative light scattering detector (HPLC-ELSD) and HPLC with UV/Visible detectors (HPLC-UV). However, it is time consumed, poor selectivity, poor sensitivity, and low applicability. Recently HPLC with tandem mass spectrometry (HPLC-MS/MS) has played increasingly important roles in the identification and quantification of triterpenoids due to its high selectivity and sensitivity. The main purpose of this study is used microwave assisted extraction to develop a HPLC-MS/MS method for analysis of ten cucurbitane-type triterpenoids (Momordicoside A, F1, F2, G, I, K, L, I aglycone, 3β,7β,25-trihydroxycucurbita-5,23(E)-dien-19-al, Momordicine I) and its application to compare the content of triterpenoids in different wild bitter melon varieties in Taiwan. In this study, microwave assisted extraction optimal conditions of extracting time 10 min, temperature 80℃. HPLC-MS/MS optimal conditions of methanol-water gradient systems with 0.01% formic acid are selected as the mobile phase of HPLC-MS/MS with the best sensitivities for triterpenoids analysis. Calibration ranges from 2 to 200 ppb with a good linearity (r>0.99) for all 10 compounds. Hulian No.5 variety has the highest content of total triterpenoids followed by Hulian No.2 and No.3 varieties. Triterpenoid content in Taiwan unique varieties (Hulian No. 1, 2, 3, 5, 6) are Momordicine Ⅰ (117.82-697.03 μg/g), Momordicoside A (21.05-412.02 μg/g), Momordicoside L (17.28-80.23 μg/g).
