Characterization of the role of phostensin in regulation of endocytic recycling

Phostensin-α and -β all are encoded by KIAA1949, but are transcribed from the different starting sites. Phostensin-α and -β consist of 165 and 613 amino acids, respectively. The sequence of phostensin-α is the C-terminal region of phostensin-β. Phostensin contains one actin-binding domain at its C-terminal region and modulates the actin dynamics. Phostensin is also associated with Eps15 homology domain (EHD) proteins. The phostensin/EHD protein complex is co-localized at the endocytic vesicles. EHD proteins have been known to play a critical role in regulation of endocytic transportation. This interaction may affect the endocytic transport mediated by EHD proteins. In my study, I have cloned the phostensin cDNA into the pmWasabi-IRES plasmid. The cell line transfected with this vector can simultaneously produce phostensin and green fluorescent protein (GFP). I carried out this system to analyze whether phostensin can regulate the endocytic recycling mediated by EHD proteins by flow cytometry
