The application of microbubble combined with pulsed laser for transdermal drug delivery

Transdermal drug delivery (TDD) has been demonstrated by ultrasound (US)-mediated microbubbles (MBs) cavitation. The present study investigated a new treatment model for evaluating the laser-mediated MBs cavitation to enhance TDD both in vitro and in vivo. The in vitro experiments show that the penetration depths in laser combined with MBs group increased 37% than control group in pigskin. In Franz diffusion experiments performed in vitro, the release rates of arbutin at 24 hours in the laser combined with MBs group resulted in 43% more than control group. Experimental animals were randomly divided into the following five groups:(1) no treatment (C) (2) arbutin alone (A) (3) laser combined with arbutin (L+A) (4) laser combined with saline and penetrating arbutin (L+Saline+A) (5) laser combined with MBs and penetrating arbutin (L+MBs+A).The degree of in vivo skin whitening (quantified as the luminosity index) in group L+MBs+A increased 40.8% at 11th day and 46.8% at 14th day during 20 days treatment. The results obtained in this study indicate that combining laser with MBs can increase skin permeability and enhance the delivery of arbutin to inhibit melanogenesis, without damaging the skin in mice.
