In this study, we had designed and manufactured a highly efficient filling microfluidic device that can be used for cell culture, polymerase chain reaction and drug screening. Precision machining was the main process of the device fabrication. The commercially available Solid Works software was adopted for the structure design. The UG software was then used to simulate the machining process. The simulated tooling file was then loaded into the CNC milling machine for mold production. The fabricated metal mold was used for pouring PDMS to obtain highly efficient filling microfluidic structures. Finally, plasma assisting packaging was conducted to tightly bond the PDMS microfluidic structure with the glass substrate. Experimental results showed that the additional semicircular filling structure and the expending fill entry structure could efficiently enhance the filling efficiency of the microchannel device. A multi-well structure could have a filling capacity of up to 904 L, requiring only a relatively short filling time of 10 min. Furthermore, the proposed highly efficient filling microfluidic device possesses advantages such as being feasible of mass production and cost effective.
