Effects of Germination Conditions on Bioactive Compounds of Brown Rice and Apoptosis of Human Colon Cell

Popis: | 105 This study investigates bioactive compounds and colon cell apoptosis of ungerminated brown rice ( UBR ) and germinated brown rice ( GBR ). We used two rice cultivars ( Oryza sativa L ), Taiwan Japonica 9 ( TJ-9 ) and Taichung Indica 10 ( TCI-10 ), as the materials in our experiments. The conditions and variables in inducing germination are: soaking time in solutions of CaCl2, Glutamic acid or RO water, for periods of 12, 24, 36 or 48 h; incubation at 30℃ in dark closed vessels. We found GBR generated much higher levels of bioactive compounds than ungerminated brown rice ( UBR ). Also we obtained the following results: the α-amylase activity of TCR-48 sample was the highest at 9.06 U/g. The GABA content of JCG-48, TCC-48, THG-48 and THR-48 samples were 47.92 mg/100g, 48.47 mg/100g, 48.47 mg/100g and 48.39 mg/100g, respectively. The γ-Oryzanols content of JCC-48, THG-48, THR-48 samples were 4528 mg/100g, 4273 mg/100g and 4443 mg/100g, respectively. The α-Tocopherol content of THG-48 sample was the highest at 0.85 mg/100g. The γ-Tocotrienol content of TCG-48 and TCR-48 samples were the highest at 3.76 mg/100g and 3.91 mg/100g, respectively. Our results show that long soaking time ( 48h ) is conducive to maximize bioactive compounds. Therefore, we chose used the samples from this condition ( 48h ) to be 1) extracted with ethanol ( E ), 2) hydrolyzed in simulated gastrointestinal situation after cooking ( CPP ) and or 3) extracted by ethanol after cooking ( CE ), then respectively, before evaluate the colon cancer cell assay, respectively. We found the highest lowest viability of the colon cancer cells was 14.2 % by using THG-48-E sample. Moreover, THG-48-E ( 150 μg/mL ) sample could arrest the G0/G1 phase of colon cancer cell cycle in G0/G1 and increase extend sub-G1 phase. Our experiments confirmed the TCI10, that after soaking time in Glutamic acid on for 48 h, temperature at 30℃, incubation in darkness and closed vessels, to with ethanol extraction, which can promote the apoptosis in of colon cancer cells. These results are considered very useful research references for the future development of functional health foods and additives. |
