Neural differentiation of neurospheres on nanofiber conduit by Schwann cells cultured medium induction

Popis: | 104 Stem cells from children exfoliated deciduous teeth with multiple differentiation capacity can be used for tissue engineering of cell replacement source. Schwann cells were the main structure and function of cells that make up the peripheral nerves. It can release of neurotrophic factors. In this study, we evaluated that we use neurosphere to differentiation into neuron then culture in conduit. After that we can observe the cells difference cell type in conduit. In our study, SHED cultured in polydimethylsiloxane (PDMS) to form neurosphere and collected down into tissue culture plate. Then we use the cells to reculture in conduit ( Random & Align ). We measured neural differentiation capacity by qPCR, and detected neuronal differentiation associated gene (Nestin, β-III tubulin, GFAP, γ-enolase). The morphology of cells were observed by using confocal microscopy as well as immunocytochemistry of Nestin, β-III tubulin, GFAP and γ-enolase measured. Based the results, in 3 days the cells started some synaptic growth on conduit. Till to 7 days it had more significant and longer synapses that can prove the induction of conduit. But between the differentiation of Align and Random conduits results. In Align conduit, cells synapse tend to growth in one way direction and no more could be observe from the neuron. In Random conduit, cells synapse tend to growth in every direction. This results could be effective in helping to nerve cell differentiation, and may be a new therapeutic for nerve damage repair. |
