Nanodiamond for theranostic applications: Toxicity evaluations and biolabeling

Nanodiamond (ND) has recently attracted great attention for its intrinsic luminescence in the wide range which can be used as a tracking marker in many biological applications. In this study, the biocompatibility and bio labeling of ND are demonstrated via the interaction with cells, microorganisms and Zebrafish embryo in vitro and in vivo. Studying ND interaction with cell we analyze the interaction of ND with Red Blood Cells (RBCs), demonstrating ND effect on RBCs oxygenation and deoxygention process. For the microorganisms, their living functions in the presence of ND are analyzed. The conditions of ND non-toxicity are discussed with varying ND size and surface properties. For Zebrafish embryo, we use the embryo model for in vivo nanotoxicity test. We optimize the conditions for using this animal model and analyze the zebrafish embryonic development in the presence of ND. ND is observed in the embryo in vivo using laser confocal fluorescence microscopy and fluorescence lifetime imaging. The origin of ND fluorescence are more than 500 luminescent defects and admixtures centers in the diamond lattice. The most studied and used are NV0 and NV- centers, mostly used in study above. Additionally, we present an experimental investigation of luminescent of Ni-related defect that emits near 885 nm in NDs. The fluorescence mapping of distribution 500 nm ND in Baby Hamster Kidney Cells (BHK) using emission of Ni-related center shows no photo bleaching and no damage to the BHK cells, the position of ND is clearly observed relatively the cell. This study is important for the bio and medical applications of ND used as a bio compatible label. Our results demonstrated nanodiamond can be used in bio imaging and drug delivery. Correspondingly, it opens further opportunities for the ND bio applications.
