Analysis of optically anisotropic properties in ovarian cancer cell and stretching biological tissue by using two-dimensional full-field ellipsometry

This study develops a new type of experimental scheme to extract optical properties by Stoke vector and Mueller matrix on biotissue and cancer cell. Using CCD camera for a two-dimensional measurement and applying differential matrix model to decouple and analyze the optical properties including linear birefringence (LB), linear dichroism (LD), circular birefringence (CB), circular dichroism (CD) and depolarization (Dep) properties of turbid media is introduced. Different from previous optical models, this system is to use two-dimensional full-field ellipsometry to measure anisotropic optical sample. It is noted that previous optical model such as photoelasticity model usually ignores the other properties on anisotropic biotissue. In this research, we have two parts for research. In the first part, the cervical and ovarian cancer cells with different chemoresistance is observed in macroscopic and microscopic images. Measuring the different optical properties between the ovarian cancer cells with different chemoresistance is to figure out how those differences come from by the macroscopic and microscopic images. This study is focus on the optical properties of cancer cells with different chemoresistance and this part has never been done before, and we hope that this technique can be applied on clinical treatments to distinguish the chemoresistance cancer cells. In the second part, we combine two-dimensional optical system and a home-made stretching machine for conducting the experiment on the biological muscle. The chicken breast muscle is chosen to figure out how the optical properties change on the stretching biological tissue. After experiments, we build up a series of simulations to observe how linear dichroism (LD), circular birefringence (CB), circular dichroism (CD) and o depolarization (Dep) affect the linear birefringence (LB) if only the photoelasticity model is applied without considering the other existing optical properties.
