Popis: | 103 In this thesis work, the pump-probe technique is further developed to take advantages of the coherence of stimulated emission, which was shown to enable long working distance detection of fluorescence. The thesis work is categorized into two parts. In the first part, the delay control between the pump and the probe pulses is improved from adjusting the length of the synchronization coaxial cable to the use of high precision digital delay generator. The timing precision and the ease of use are thus greatly enhanced, which allows quick and precise measurement of fluorescence lifetime. In the second part, the Michelson interferometry is integrated into the pump-probe setup for coherence gating to realize axial resolution. In this way, molecular sensitivity can be brought into optical coherent tomography (OCT), which may possess immense application potentials for optical biomedical imaging. |
