Column is an important part in the separation techniques (Electrophoresis, Liquid Chromatography, High Performance Liquid Chromatography) in recently. Main function of the Column is to separate (purified) the sample. The principle is make use of different affinity of sample cause to the different retard time to separation. Its structure and material will affect the separation efficiency. The most literature of miniaturized column is designed to the circular pillar array in the current, and the majority of the material is silicon-based or polymer-based. Silicon-based column is the major direction of the research. The aspect ratio can be up to 40 and apply to high-performance liquid chromatography. The characterized of Polymer-based column can be made quickly by the reprographic technology, but often encounter the problem of inadequate Young's modulus will be fall. The research focus to make of high aspect ratio polymer-based column and test its separation efficiency. Hoping the aspect ratio of polymer column can be made as the same as silicon-based column, and shorten the separation time to achieve the purpose of rapid detection. This study will compare separation sufficiency with silicon-based, polymer-based and commercial column. From the results, this experiment can successfully create a PDMS column aspect ratio about 32, and silicon column aspect ratio about 31.Both column are successfully separate the sample of benzene and benzophenone. Silicon polymer The separation time is only about 1.5 minutes for Silicon-based and polymer-based column. The separation time of commercial column is less than above.
