Capillary Electrophoresis Coupled with Multiple Detection Methods for Simultaneous Analysis of Alendronate, Pamidronate, Ibandronate and Zoledronate in Human Plasma

Alendronate, Pamidronate, Ibandronate and Zoledronate are currently the four most widely used bisphosphonate drugs for clinical treatment of osteoporosis and fractures caused by cancer. Bisphosphonates can inhibit osteoclast activity, which slows bone loss. Most bisphosphonates do not possess UV absorbing or fluorescent functional groups, and therefore they are not easily to be analyzed. This thesis describes the development of a capillary electrophoresis method coupled with UV absorption detection, fluorescence detection and electrochemiluminescence detection for simultaneous analysis of four bisphosphonates. The obtained detection limits for Alendronate, Pamidronate, Ibandronate and Zoledronate are 10 ppb, 20 ppb, 30 ppb and 0.87 ppm, respectively. Without preconcentration, the sensitivity of the method was not adequate for real sample analysis. Solid phase extraction using Fe3O4@Al2O3 magnetic nanoparticles as extractant was employed to preconcentrate the drugs before analysis. The detection limits for Alendronate, Pamidronate, Ibandronate and Zoledronate in plasma through solid phase extraction can be further lowered to 3.6 ppb, 4.2 ppb, 9.2 ppb and 0.51 ppm, respectively. Using Fe3O4@Al2O3 magnetic nanoparticles as extractant in solid phase extraction not only effectively reduce the detection limits of analytes but also eliminate the complex plasma sample matrix. Electrophoretic separation of the four drugs can be completed in six minutes. Including the procedure of solid phase extraction, the whole analysis can be finished within one hour. This developed method is simple, rapid, convenient and highly sensitive.
