Dendrobium is a major floral crop around the world as a cut flower, medicinal plants, breeding, or ornamental plants. The present study had been completed to observe the seed germination and plantlets growth in medium containing different concentration of organic nitrogen, organic additives, activated charcoal in in vitro, and seedling growth in the green house. Liquid culture is suitable for germination, it also germinated faster than solid medium. On the other experiment, ? MS solid medium got the highest percentage compared to another media. In this study proved that organic nitrogen, tryptone has the highest value of height compared to peptone and casein. In the organic additive, coconut water give the best result for plantlets growth due to it has a more complete nutritions. activated charcoal has an important role in tissue culture for plant growth. This can be seen in the result that the best concentration for plant growth plantlet culture of Dendrobium victoria-reginae Loher var. miyakei is 2 g L-1 activated charcoal. Seedling growth is an important factor after culture in in vitro. Their initial growth in in vitro being followed by transplanting into green house or field. Based on this study, 0.5 g L-1 Hyponex-2 give the best result for the stem growth of Dendrobium victoria-reginae var. miyakei in green house.
