Liposomes uptake by the cell membrane model

In the recent years, life can be extened within medicine development, but still there are some diseases couldn’t be cure like cancer. Cancer treatment are not easy and may follow with some serious side effects. Therefore developing new medicine or new dosage forms becomes more important. Liposome is a kind of new dosage forms, because of it’s structure and chemical composition make it easy uptake by body and also release slowly. Using ITO glass electro-swelling forming liposomes with diameter more than 50μm as model of cell membrane (GUV). Extrusion 200 nm diameter liposome to carry drug. Mixture of lipid with DOPC:DPPC:CHOL(2:2:1) added 5% NBD GUVs, used fluorescence microscopy identify intense CHOL membrane domain on the GUV. Loading with four kind of SUV : pure DOPC、 DOPC:Chol(7:3)、pure DMPC、DMPC:Chol(7:3) both with 5% Hypericin. Observing SUVs truly uptaking by GUVs, and SUVs .Both stay on the intense CHOL membrane domain. Modify GUV chemical composition DOPC、DPPC CHOL 2:2:1 to 4:4:1 and 1:1:1, loading with DOPC:Chol(7:3). By decreasing GUV Cholesterol concentration SUV can be found on intense chol membrane domain, and will more easy uptake by GUVs.
