Popis: | 101 We firstly synthesized functionalized carboxymethyl chitosan(CMCS) by carboxylation reaction of chitosan (CS), and then degraded CMCS by peroxide to form low-molecular weight carboxymethyl chitosan (LMCMCS).These materials were used to modify screen-printed electrodes(SPEs) for preparing an inexpensive, disposable glucose biosensor with glucose oxidase and ferricyanide (as a mediator). It was foundthat the viscosity average molecular weightof the degraded CMCS decreases as the reaction time uncreases. Besides, we used scanning electronicmicroscopy to observe the micro-structure of the SPE surfaces. And then the electrochemical behaviors of various modified SPEswere investigated by CV andEIS.Results show that the LMCMCS-modified SPE demonstrates not only better electrochemical characteristics but a significant enhancement of catalytic current for glucose. Moreover, the electrochemical behavior of the modified SPE belongs to diffusion-control kinetics. The optimal operation conditions of glucose biosensor, such as working potential, enzyme loading, and pH were 0.3V, 0.8U and pH 7,respectively. Moreover, the sensitivity of the resulting glucose biosensor is ca. 0.545 μA/mM, which increases over 23.6 % comparing to bare one, with a wide linearity up to ~33 mM. |
