Safety evaluation on the ethanolic extracts of Fomitopsis pinicola

Fomitopsis pinicola has been traditionally used as an antidiabetic material in Korean folk medicine and is known to be a nontoxic mushroom in Europe; a number of its different principles such as triterpenes, steroids, and a heterogalactan have been shown to exert medicinal effects, including antimicrobial action and the inhibition of cyclooxygenase activity. Safety evaluation assays were performed: The Ames test of Salmonella typhimurium tester strains(in vitro); and micronucleus assays for mice (in vivo). Acute oral toxicity and 28-day repeated feeding toxicity tests were performed by oral gavage in wistar rats. Fomitopsis pinicola did not increase micronucleus ratios in vivo, Fomitopsis pinicola had no acute lethal effect at the maximum dose (5 g/kg/day bw) in rats. In the 28-day study, there were no adverse effects on body weight, feed consumption, hematology, blood biochemical parameters, organ weight, or pathology. The acute toxicity study showed that the LD50 of Fomitopsis pinicola was greater than the tested dose (up to 1 g/kg/day bw) in wistar rats. In the subacute toxicity study, the no observed adverse effect level (NOAEL) of FP in rats was 1 g/kg/day bw. The in vivo study of mammalian erythrocyte micronuclei confirmed the Ames test results, demonstrating that FP has no mutagenicity.
