In this study, we used polymerase chain reaction - denaturing gradient gel electrophoresis ( PCR-DGGE) and bacterial 16S rDNA sequencing to analyze the gastrointestinal tract(GI tract) bacterial diversity of Giant panda. The experiment started from 2009 May to 2010 May. PCR-DGGE DNA fragments of different strains in the sample separated in the DGGE gel, the obtained electrophoresis result makes similarity of analysis again the V1~V3 variation area nucleic acid banding distribution by Phortix 1D. The result discovered in feeds ingredient stable period, the V1~V3 region fragment similarity respectively is 72% and 71%. In feed invariable period, The GI tract microbiota of giant panda presents the steady state, but the same time, similarity between the GI tract microbiota of the female pandas and the male panda is 55%. The GI tract microbiota of the female panda is difference with the GI tract microbiota of the male panda. Moreover, we also compared the GI tract bacterial diversity in oestrus with the GI tract bacterial diversity in non-oestrus. The result discovered that the similarity decreased from 71% to 45%. That expressed the change of the GI tract bacterial diversity because of the oestrus. We analyzed the GI tract microbiota 16S rDNA fragment sequence, discovered that the major strains are Enterobacter spp.、Klebsiella spp.、Pseudomonas spp. and Hafnia alvei. Using Community level physiological profiling analyzed the metabolic capacity of anaerobic bacteria in the intestinal tract in the gaint panda. The experimental result discovered the metabolic capacity of anaerobic bacteria using sugar in September and December better than in June. But the metabolic capacity of anaerobic bacteria using acid in Jane is better than in September and December. The reason is the different of the count of GI tract microorganism between in summer to in winter. The count of GI tract microorganism is less than in winter. The energy source of panda is carbohydrate. Because the GI tract of panda is not good for obligate anaerobe, the facultative anaerobic organism is the major group in the GI tract of panda. When the count of GI tract microorganism decreased, we can discover the obligate anaerobe.
