A Proteomic Analysis of PC12 Cells with Low-Intensity Ultrasound Stimulation

Low-intensity ultrasound (LIUS) has been reported with the ability to accelerate the regeneration of peripheral nerve tissues, to affect cells’ differentiation and proliferation and to induce the apoptosis of cancer cell lines by regulating genes and protein expression.1,2 However, detailed protein expression after LIUS stimulation has rarely been analyzed. Therefore, we try to use the mass spectrometry (MS) to analyze the proteome of cells after the stimulation of LIUS. In this study, LIUS was used to stimulate PC12 cells, a neuron-like cell line derived from a pheochromocytoma of the rat adrenal medulla. The in-gel digestion method was used to prepare samples for the bottom-up proteomic analysis. In this work, we used liquid chromatography (LC) coupled with the MS to determine the mass of peptides and then used the Maxquant software to identify proteins. We identified 3087 proteins in the control group and 3101 proteins in the experimental group and 65 proteins had clear change after the stimulation. In these 65 proteins, some proteins are related to neurite growth, membrane trafficking, stress response and cell migration. Hence, the results indicate that LIUS may influence cell development but the detailed mechanisms still need to be further examined.
