Bipolar disorder shows numerous dysfunctions in emotion, motor, and cognition. Moreover, its animal model remains uncertainty. The purposes of the present study examined issues as below. (a). In the study, administration of ouabain (Na+/K+ ATPase inhibitors) to induce the symptoms of bipolar disorder to assess the locomotor and spatial learning performance using the Morris water maze test. Further, it was established to ouabain-induced bipolar disorder for an animal model. (b). The typical anti-bipolar disorder drugs such as lithium, carbamazepine, and valproate has been used to test their effects for bipolar disorder in the spatial learning. (c). The concentration of corticosterone in plasma was assessed after the locomotor test. (d). The extracellular signal-regulated kinase-2 was measured to the medial prefrontal cortex-amygdala connectivity and hypothalamus after the Morris water maze test. The present results showed that: First, ouabain induced the hypoactivity in the locomotor and impairments of the congnitive performance. Second, the typical treatment drugs (such as lithium, carbemazepine, and valproate) that has different therapeutic effects in the rat of bipolar disorder. However, the Lithium treatment is the most available drug for anti-bipolar disorder. Third, ouabain-treated rats showed to be non-significant differences in concentration levels in plasma compared to the control. Finally, the extracellular signal-regulated kinase-2 has significantly lower only in the amygdala. The present findings should be discussed and it can provide some insights for clinic.
