Dynamical Fluorescent Imaging of Biological Cells

We constructed a spectral imaging system for observation of fluorescent labeled biological cells. The system is based on an inverted fluorescence microscope and can perform both of wide-field imaging and scanning fluorescent imaging. In the system, biological cells were cultured in a temperature- and humidity-controlled incubator which was integrated in the microscope such that the system was able to perform the observation with time-lapsed imaging in a long period. Furthermore, we proposed an algorithm and an automatically processed program for contrast enhancement and background removing of images. After image processing, the contrast of images was significantly enhanced and the fine structure of biological cells can be observed. The lateral resolution of the system was examined to be 5.52 μm. This system is capable to observe biological cells labeled with multiple fluorescent markers. We also proposed an algorithm based on the principle of iteration such that the distribution of fluorescent markers can be analyzed quantitatively.
