The study of hylauronan conjugated gold nanoparticles for the uptake of cells with CD44 receptor

Popis: | 101 The aim of this research is to study the surface modification of hyaluronan (HA) for Au nanoparticles (AuNPs) on the endocytosis of cells with CD 44 receptors. Two cell lines MDA-MB-435s (with CD44) and MDA-MB-453 (without CD44) co-cultured media with/without AuNPs are investigated for experiments. The qualitative observations are conducted using dark-field microscopy and laser scanning confocal microscopy. In addition, the quantitative measurements using ICP-OES are provided for comparison. Moreover, the specific conjunction of the ligand and receptor is further studied for the application of cellular photodamage. The surface modification of covalent bonding for AuNPs with HA is confirmed by using 1H-NMR spectroscopy and zeta potential. Our results show that within the same incubation time the uptake dosage of AuNPs modified with HA by MDA-MB-435s is higher than that without HA. This implies that the receptor-mediated endocytosis can speed up the cellular uptake of AuNPs. In addition, under the exposure of a pulsed laser (532 nm) the cell morphologies also show that the cellular photodamage of the former is severer than that of the latter. |
