Quantitative measurement of protein expression is the most common method for cancer detection. The performance of current serum markers, however, is dissatisfactory. Genetic variations and post-translational modifications of proteins have been reported to be associated with disease pathogenesis. Therefore, variant pattern of cancer-specific proteins may provide useful information in disease discrimination. In this study, we aimed at verifying whether or not the protein variant pattern is a better marker candidate than the expression level of a single protein. Taking serum amyloid A as a model protein, we combined the NBAMS assay and bioinformatics approaches to evaluate the feasibility of utilizing protein pattern as potential markers of diseases, in this study, gastric cancer and colorectal cancer.
