The Study of p-Phenylenediamine and m-Aminophenol Hair Dyes on Melanogenesis and Keratinocytes Damage

Hair dyes is getting more popular to upgrade appearance of people because of its convenient and fashion. Many reports indicated that those dyes induced allergy and inflammation response, so it’s important to be safe. In this study, we use p-Phenylenediamine (p-PD), m-aminophenol (m-AP), Hydrogen peroxide (H2O2) and the mixture of those compounds (Mixture) to analyze the influences of melanogenesis and damage of keratinocytes. The results shows that the IC50 values of cell viability for B16-F10 are 284.8±2.5 μM (p-PD, 24 h), 5.0±0.1 mM (m-AP, 24 h), 476.9±1.9 μM (H2O2, 24 h), 395.2±1.6 μM (Mixture, 24 h), 33.7±0.4 mM (p-PD, 30 min), 6.7±0.4 mM (H2O2, 30 min) and 1.9±0.1 mM (Mixture, 30 min), respectively. There is no cytotoxicity to B16-F10 cell line if the concentrations of p-PD, m-AP, H2O2 and Mixture are lower than 50 μM (p-PD, 24 h), 1 mM (m-AP, 24 h), 15 μM (H2O2, 24 h), 50 μM (Mixture, 24 h), 15 mM (p-PD, 30 min), 4 mM (m-AP, 30 min), 0.1 mM (H2O2, 30 min) and 0.1 mM (Mixture, 30 min). Moreover, these compounds can decrease 33.5 % (p-PD, 24 h), 53.0 % (m-AP, 24 h), 27.6 % (Mixture, 24 h), 39.8.0 % (p-PD, 30 min), 18.4 % (m-AP, 30 min) and 8.1 % (Mixture, 30 min) of the melanin content, respectively. Mixture shows that which was not decrease cell viability of XB-2 cell line when the concentrations were less than 100 μM (24 h) and 0.25 mM (30 min), and cytotoxicity was 17.1 % at 550 μM. As the concentration was 1 mM, it can increase 2.04-fold of caspase-3 activity and 4.57 % of DNA fragmentation at 36 and 24 h when compared with control. The results indicated that p-PD, m-AP and Mixture can reduce the formation of melanin in B16-F10 cell and Mixture can induce apoptosis in keratinocytes.
