To Analysis the Immune Functions in Hemodialysis Patients

Several studies have been made on the immune functions in hemodialysis patients. It has been reported that T cells dysfunction and Genetic polymorphism of cytokines (included Cytotoxic T –Lymphocyte Antigen 4; CTLA-4, Interferon-gamma;INF-γ, Interleukin-6; IL-6, Interleukin-10;IL-10, Interleukin-12;IL-12, Transforming growth factor-β; TGF-β) in hemodialysis patients, which result in reduced the body's ability to defend against pathogens, thus increasing the risk of infection with hepatitis B and other diseases. Blood samples of hemodialysis patients with (male 23, female 32) or without (male 13, female10) Hepatitis virus B infection were obtained from Pingtung Christian Hospital. The peripheral blood mononuclear cells (PBMC) were separated by Fill-hypaque and stained with CD4+-FITC and CD8+-PE to analysis the CD4+/CD8+ ratio by flow cytometry. Next, the MOLT4 cell line were co-culture with patient’s serum to analysis the apoptosis effects of patient’s serums. In addition, the polymorphism of, CTLA-4 (+318T/C、+49G/A), INF-γ (+874A→T), IL-10 (-1082G/A)、IL-12(+1188A/C)、TGF-β(+869C/T、+915G/C) and will be analyzed by ARMS-PCR. The results indicated that there was significant difference on CD4+/CD8+ ratio between dialysis and non-dialysis patients. In the apoptosis analysis, the result indicated that there was significant difference between dialysis and non-dialysis patients.
