The Gene Expression of Insulin-like Growth Factors in Gonad during Sex Differentiation of Orange-spotted Grouper, Epinephelus coioides

Popis: | 99 The purpose of this study was to investigate the gene expresstion of insulin-like growth factors (IGFs) and their receptors during the sex differentiation of orange-spotted grouper (Epinephelus coioides).Gonadal sex differentiation in orange-spotted grouper occurred around 120 to 150 days after hatching (dah). To investigate the role of IGFs during the gonadal sex differentiation and development, we analyzed the IGFs gene expression of the orange-spotted grouper from 72 dah to 515 dah (about 1.5 years). The expression of igf-1, igf-1r and igf-2 increased during gonadal sex differentiation, and insr, vasa and nanos were expressed increasingly during the gonadal development. insr and igf-2r were expressed at higher level in 72 dah. These results suggest that IGFs, vasa and nanos are associated with the goandal sex differentiation and development. The grouper treated with 1,4,6-androstatriene-3,17-dione (ATD) before the sex differentiation were attenuated at the expression of IGFs, vasa and nanos, and thereby affecting the gonadal development. The grouper treated with estradiol-17β (E2) were attenuated at the expression of IGFs and nanos but enhanced at the expression of vasa. By immunohistochemistry, insr was expressed in some somatic cells of gonad during the gonadal development. igf-1r and insr were expressed in the follicle cells surrounding the previtellogenic oocytes during the oocyte development. vasa and nanos were expressed specifically in germ cells. By immunofluorescence, insr was expressed in some somatic cells of the gonad during the gonadal development. insr, vasa and nanos were colocalized in few primary oocytes during the gonadal development. |
