Identification and functional analysis of the components stimulating pollen germination in lily stigma/style exudates

Pollen germination and tube growth are critical processes for successful double fertilization in higher plant. After landing on the stigmatic surface, dehydrated pollen grains will receive female signals to trigger pollen hydration, activation and pollen tube emergence. Many factors present in the stigma/style exudates have been suggested to be involved this sophisticated program, but very limited information was known about their composition and the identities of the active molecules involved in pollen-stigma interaction. In this thesis, first we examined the effect of lily stigma/style exudates on lily pollen germination and tube growth; followed by isolation and characterization of the composition of exudates. Our results clearly showed that stigma/style exudates significantly promoted pollen germination rate and the active molecules for germination enhancement were heat- and proteinase K-resistance. The later results suggested that the pollen germination promoting molecule is probably not protein. Then, we extracted the stigma/style exudates by a series of different solvents (ethyl acetate, n-Butanol and water) partition to separate the potential active molecules. Interestingly, molecules present in the layer of ethyl acetate even showed more significantly enhanced effect on pollen germination rate than those in the lily stigma/style exudates did. Then we use NMR、GC-MS、LC-MS and ESI Q-TOF MS analysis layer of ethyl acetate, but they can not be a good report. In addition, two-dimensional PAGE were used to reveal the total protein profiles of germinating pollen affected by the active molecules in the layer of ethyl acetate and the identities of several protein spots were determined by LC-MS/MS. We find 13 protein spot in the layer of ethyl acetate, these spot maybe in amino acid metabolism and signal translation for enhance pollen tube germination.
