Studies on isolation of alginate lyase-producing Bacillus megaterium strain from soli and characterization of the lyase.

In this study, 156 bacteria have been isolated from soil and decayed brown algae. Strain P44 showed the strongest alginate lyase activity among isolates which can use alginate as sole carbon source. After gram staining, spore staining. catalase test, API 50CHB identification and 16S rRNA sequencing, P44 identified as Bacillus megaterium. Bacillus megaterium P44 can break down poy-1,4-D-mannuronate and poly-1,4-L-guluronate in the alginate polymer, produce protease and catalase, and cannot use CMC and agar as carbon source. After incubation at 37℃, 120 rpm for 72 to 86 hours in the enzyme production medium, Bacillus megaterium P44 is able to produce alginate lyase of 386.4 U / ml, the enzyme optimum temperature and pH values are 45℃, pH5.5 ~ 6.5, and the optimal stability range should be 35℃, pH 5.5 to 6. Bacillus megaterium P44 can produce alginate lyase 29 U / ml in Sargasso medium which use 2% Sargasso powder as the sole carbon source.
