Development of Sequence Characterized Amplified Region (SCAR) markers associated with pepper anthracnose (Colletotrichum acutatum) resistance

Popis: | 99 Anthracnose (Colletotrichum spp.) is one of the most damaging diseases of pepper during the warm/wet season. The main objective of this study is to identify AFLP molecular markers associated with pepper anthracnose resistance and conversion to SCAR markers. F2 population derived from cross of the resistant parent CCA0038-9155 and the susceptible parent PP9955-15 was assayed for resistance at the green mature stage. Out of a total of 772 polymorphic bands generated (317 from PstI/MseI digestion and 455 from EcoRI/MseI digestion), two AFLPs (Amplified fragment length polymorphism) (A257 and A518) demonstrated strong association with anthracnose resistance at green mature stage. AFLP marker A518 associated with pepper anthracnose resistance was cloned and sequenced. An insertion/deletion (INDEL) was identified between resistant and susceptible genotypes. A primer pair was designed for a SCAR (sequence characterized amplified region) marker assay utilizing the 10-bp INDEL. Based on the results, this INDEL marker is highly correlated with the original AFLP marker A518 and anthracnose resistance at the green mature stage. Thus, this marker should be effective in marker-assisted selection of pepper progenies derived from PBC932 for resistance to anthracnose. |
