Effect of hypoxia-induced factor on insulin sensitivity in skeletal muscle cells

Popis: | 99 Insulin resistance is a risk factor of several chronic metabolic diseases. In the previous studies, exercise has been shown to exert beneficial effects on insulin sensitivity. Exercise may cause hypoxic phenomenon in human body, and hypoxia-induced factor-1(HIF-1) can be induced under hypoxic condition. In the present study, we tried to verify the association between hypoxia- induced HIF-1 and insulin sensitivity in differentiated C2C12 myotubes. After 6 hr incubation under hypoxic condition, expression of HIF-1α, insulin signaling pathway, and insulin-induced glucose uptake were analyzed. A HIF-1 inhibitor, YC-1, was used to investigate the role of HIF-1 on insulin-stimulated glucose uptake, and insulin signaling pathway. Our results showed that hypoxia increases HIF-1α protein expression, insulin-induced glucose uptake, and insulin-stimulated Akt phosphorylation in C2C12 myotubes. Inhibition of hypoxia-induced HIF-1 by YC-1 treatment suppressed insulin-induced glucose uptake, but insulin-stimulated Akt phorphorylation was not affected by YC-1 treatment. In conclusion, the HIF-1-regulated insulin-stimulated glucose uptake is mediated by an Akt-independent pathway under hypoxic condition. |
