Column Purification and Anti-solvent Recrystallization of Zeaxanthin dipalmitate from Lycium barbarum and Effect on Proliferation of Adult Retinal Pigment Epithelium Cells

Popis: | 99 This study employed ultrasonic extraction, column elution fractionation and supercritical carbon dioxide anti-solvent recrystallization in preparing the pured nano-sized zeaxanthin dipalmitate (ZDP) from Lycium barbarum fruits. Column fractionation of ultrasonic acetone extract coupled with supercritical carbondioxide anti-ethyl acetate solution ehanced amount of ZDP from 72.75 mg/g in the acetone extract to 931.5 mg/g in the SAS precipitates. The ZDP recovered at 25.2% and 64.9 % from Lycium barbarum fruits individually by liquid anti-solvent and by supercritical anti-solvent recrystallization. Experimental design by response surface method showed that feed concentration and feed flow rate are important on the purification and the preparation of leaf type nano-sized ZDP containing particles. These purified ZDP precipitates in aqueous dimethyl sulfoxide solution exhibited anti-oxidative abilities on proliferating human adult retinal pigment epithelial cells (viability from 22% to 31%), though it was a meager increase compared to those of zeaxanthin (viability from 43% to 94%). |
