Studies on the chemical constituents and biological activities of the stems of Ludwigia octovalvis (Jacq.) Raven

Popis: | 98 Ludwigia octovalvis (Jacq.) Raven (Onagraceae), an aquatic plant, is widely distributed in wet areas of Taiwan. It is used as a traditional treatment for edema, nephritis, and hypertension. In this study, the air-dried stems of L. octovalvis were extracted with methanol, which was then partitioned to give n-hexane, ethyl acetate (EtOAc), n-butanol (n-BuOH), and water layers. In the bioassays, the EtOAc layer of the stems of L. octovalvis showed antioxidant and anticancer activities. Therefore, the EtOAc layer was further purified by column chromatography, HPLC, and recrystallization to obtain sixty-four compounds, including seventeen triterpenoids, twenty-two aromatics, ten steroids, two lignans, six apocarotenoids, five phenolic lactones, one alkaloids, and one benzoquinones. Their structure were identified on the basis of spectroscopic methods. Among these, 2α,3β,19-trihydroxy-23-trans-ferulyloxyurs-12-en-28-oic acid (T17), erythro-2,3-bis-(4-hydroxy-3-methoxyphenyl)-3-methoxypropanol (Ar20), (+)-erythro-2-[2-hydroxy-5-(3-oxo-1-propenyl)-3-methoxyphenyl]-1-(3-hydroxyl-5-methoxyphenyl)-propane-1,3-diol (Ar21), and (+)-erythro-2-[2-hydroxy-5-(3-oxo-1-propenyl)-3-methoxyphenyl]-1-(4-hydroxyl-3,5-methoxyphenyl)-propane-1,3-diol (Ar22) are new compounds. The pure compounds, betulinic acid (T1), betulonic acid (T2), 3-acetylbetulinic acid (T3), lawsonic acid (T4), 23-trans-p-coumaroyloxy-2α,3β-dihydroxyolean-12-en-28-oic acid (T12), methyl gallate (Ar6), (E)-coniferaldehyde (Ar9), (+)-erythro-2-[2-hydroxy-5-(3-oxo-1-propenyl)-3-methoxyphenyl]-1-(3-hydroxyl-5-methoxyphenyl)-propane-1,3-diol (Ar21), (+)-erythro-2-[2-hydroxy-5-(3-oxo-1-propenyl)-3-methoxyphenyl]-1-(4-hydroxyl-3,5-methoxyphenyl)-propane-1,3-diol (Ar22), 4-ketopinoresinol (L2) and 2,6-dimethoxy-1,4-benzoquinone (Be1) showed significant cytotoxic activies against SK-Hep1 cell line with IC50 values of 6.9, 38.0, 17.3, < 4.9, 16.2, 58.2, 33.8, 38.5, 38.7, 9.7 and < 18.6 μM, respectively. Furthermore, syringic acid (Ar4), gallic acid (Ar5), methyl gallate (Ar6), (E)-ferulic acid (Ar8), (E)-coniferaldehyde (Ar9), and syringaresinol (L1) showed significant DPPH free radical scavenging activities with the EC50 values of 60.7, 30.5, 62.2, 90.4, 114.1, and 42.5 μM, respectively. In addition, the n-BuOH layer showed protective effects on tert-butyl hydroperoxide (t-BHP)-induced hepatotoxicity. |
