Study on ABRC3-CBF1 gene transformation of chrysanthemum by microprojectile bombardment

For agriculture, to breed cold-resistant, drought-resistant, and salt-resistant crops is essential. Selecting stress-resistant genes and transfering them to the crops have been an efficient technique to create stress-resistant crops. In this study, a DNA cassette containing an Arabidopsis(C-repeat/dehydration-resposive element binding factor)(CBF1)cDNA and a nos terminator, driven by a barley ABA inducible promoter, was transferred into the chrysanthemum with the method of gene gun bombing. During the gene gun bombing process, the callus bombed by 900 and 1100 psi was cultured in the medium containing 25 ppm Kanamycin for putative transgenic product selection. Using Kanamycin and GUS as early selection methods and further analyzing by PCR, we obtained 6 putative trangenic lines. Subsequently, we treat wild type and transgenic chrysanthemum with normal-temperature and low-temperature condition. The RT-PCR results show that wild type chrysanthemum did not transcript CBF1 and transgenic chrysanthemum had CBF1 transcripts only after low-temperature treatment. Nevertheless, the growth and development of putative transgenic chrysanthemum have no significant difference from wild type. Further investigations in stress-resistant ability of these transgenic chrysanthemum are needed for the future agricultural application.
