Screening of lactic acid bacteria isolated from plant resources with immunopotentiating activities on RAW264.7 macrophages

Popis: | 98 The present study aims were to investigate the immunopotentiating activities of lactic acid bacteria isolated from plant resources (LABIPR) in vitro. A murine macrophage cell line RAW264.7 was stimulated with several strains of LABIPR. Phagocytotic assay by neutral red, and the production of nitric oxide (NO), tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6) were evaluated the immunopotentiating activities of LABIPR on RAW264.7 cells. The results showed that 30 LABIPR strains could stimulate the phagocytosis of macrophages. The twelve LABIPR strains including B0018, B0019, B0020, B0040, B0042, B0044, B0046, B0052, B0088, B0091, B0110 and B0145, could stimulate the NO production of macrophages. NO was known to affect the secretion of several cytokines containing IL-6 and TNF-α. Some strains of lactic acid bacteria express significant levels of iNOS-like protein as shown by Western blot analysis.Among, this study about the significantly activation of murine macrophage Raw 264.7 cells showed that heat-killed LABIPR B0040,B0046 and B0091 stimulated the production of IL-6 and TNF-α production, nitric oxide release, and increased phagocytic activity in macrophages, express of iNOS-like protein. These results, taken together, indicate that LABIPR might play a critical role in the immunopotentiating activities. |
