The Effects of Phytoestrogens on Plasma Protein Profile of Roosters

Soybeans and soybean products are the richest identified sources of isoflavones, They have long been known for their estrogen-like effect on mammals. Many studies had provided evidence of their cholesterol reducing effects and protection against certain types of cancers, including breast cancer, prostate cancer, cardiovascular disease, osteoporosis as well as other health benefits in humans. Isoflavones are also very strong antioxidants. Previous studies on dietary intake of soy protein with isoflavones indicated that they may be associated with reductions in serum cholesterol .These findings lent supports a role for soy protein or isoflavones in reducing serum lipids. The purpose of this study is to ascertain the effects of soy isoflavones on plasma protein profiles using adult roosters as an animal model. Plasma protein were subjected to proteomic analysis that is two-dimensional PAGE and MALDI Q-TOF MS. Some proteins which are responsive to isoflavones as well as estrogen treatment have been identified and described in this thesis. The administration of 17 beta-estradiol (20 mg / kg body weight ) to growing roosters markedly increased the concentrations of plasma total protein and total cholesterol components. At the same time, levels of total cholesterol in plasma were greatly elevated. The respective values for protein and total cholesterol in plasma were 33.88 ( mg / mL ) and 153.43(mg / dL) in the zero-time control, and 45.84 ( mg / mL ) and 204.71 (mg / dL) in roosters upon estrogen treatment for three day. The results were significant (p < 0.05) in estrogen-treated roosters. The respective values for total cholesterol in plasma were 273.55 (mg / dL) in the zero-time control, and 125.07 (mg / dL) in roosters upon isoflavones treatment for twelfth days. The results were significant (p < 0.05) in isoflavones-treated roosters.
