Folate-Mediated Pluronic Modified Fe3O4 for Combined Diagnosis and Therapy

Popis: | 97 Magnetic resonance (MR) tracking of superparamagnetic iron oxide labeled cells is a relatively new technique to non-invasively determine the cancer cells. In this study, Pluronic® F127 (PF127), which is capable of encapsulating hydrophobic drug due to its PPO hydrophobic segments, was proceeded a condensation reaction with water-soluble polyacrylic acid bound iron oxide (PAAIO). A tumor-specific targeting ligand, folic acid (FA), was also grafted onto PF127-PAAIO to give FA-PF127-PAAIO magnetic micelles. A multifunctional FA-PF127-PAAIO can be used as combined diagnosis and therapy purposes specifically targeting to cancer cells. PF127-PAAIO magnetic micelles without folic acid were used as a control group. According to IR and UV absorption spectra, we ensured the successful preparation of the magnetic micelles with polymeric compartments for carrying a drug. The hysteresis curves of SQUID magnetometer analysis also showed that these magnetic micelles were superparamagnetic and a saturation magnetization was obtained in FA-PF127-PAAIO and PF127-PAAIO. Internalization of FA-PF127-PAAIO in targeted cells (KB, A549 cells) was observed by flow-cytometric analysis and in vitro MR imaging. These preliminary results demonstrated that FA-PF127-PAAIO have the ability to target folate receptor. |
