Mechanisms of regulation of GLUT4 protein expression and insulin sensitivity after prolonged aerobic exercise in rats

Popis: | 97 Purpose: Aerobic exercise has been defined as “any activity that uses large muscle groups, can be maintained continuously, and is rhythmic in nature”. It had been demonstrated that aerobic exercise can improve the control of blood glucose in hyperglycemic condition. Many different aerobic exercise regimens had been designed. In this study, we attempted to elucidate the enhancement of glucose homeostasis by a persistent daily short-term aerobic exercise regimen. Methods: The male Wistar rats were used in all experiments, which were randomly divided into the control and aerobic exercise groups. The aerobic exercise was designed that rats swim 60 min per day, 5 days per week, and persistent 6 weeks. Blood glucose, plasma insulin and triglycerides levels, oral glucose tolerance test (OGTT) were determined. In addition, the protein levels of GLUT4, AS160, AMPK, PKC and Akt of skeletal muscle for 6 weeks aerobic exercise were analysis. Result: Six weeks of aerobic exercise training improve control of blood glucose in hyperglycemic condition, fasting plasma glucose levels, insulin sensitivity, plasma triglycerides concentration, and increase skeletal muscle GLUT4 expression which through activation of AMPK and AS160 protein expression. Conclusion: Collectively, these finding suggest that six weeks of aerobic exercise training enhances control of blood glucose in hyperglycemic condition, insulin sensitivity, and increase skeletal muscle GLUT4 expression, which may contribute to an increase insulin-stimulated GLUT4 protein concentration. This study will provide important ex-induce that long-term aerobic exercise can application protective effect to improve blood glucose regulation and insulin sensitivity. |
