Study of the effects on the inhibition of Japanese encephalitis virus infection in mice brain after (-)-epigallocatechin gallate (EGCG) treatment

Popis: | 96 (-)-Epigallocatechin gallate (EGCG), a polyphenol of catechin extracted from green tea, has been investigated to have anti-viral effects. However, Japanese encephalitis virus (JEV) may cause acute encephalitis in human and damage the brain. Until now there is no effective drug for curing JE. This study investigated the inhibitory effects of EGCG and the proteomics of mice brain on JEV infection. In vitro data showed that 50 μM EGCG significantly inhibited the infection of JEV in Vero cell, and significantly decreased the mRNA expression level of iNOS and RANTES. We propose that severe inflammation responses after JEV infection would irreversibly damage the neuron of mice brain. In vivo study, C3H/HeN mice were fed with 1mg EGCG/mice per day for one month and followed with 50-fold LD50 JEV (Beijing-1 strain) challenge. In JEV infected C3H/HeN mice model, neurobehavioral abnormalities appeared at the 5th day and died at the 7th day in the post-infected mice. The survival rate of JEV infected mice fed with EGCG was raised about 20% higher than that fed with water. These results suggested that EGCG may ease the severe inflammation responses. On the other hand, we used 2-dimentional electrophoresis in conjugation with LC/MS/MS to analyze the cerebrum proteins extracted from the JEV infected mice. The data showed that thansthyretin protein (Ttr) over expressed while visinin like-1 protein (Vsnl1) was down expressed in JEV infected mice cerebrum. These proteins might play important roles in the pathogenesis of JEV infected mice brain. |
