Protective effect and mechanism of Morus alba L. on hypertensive cardiomyopathy

Popis: | 96 Morus alba. is applied for hypertension treatment in traditional Chinese medicine. Its extract contains falconoid and GABA which has been determined with the efficacy of antioxidant and blood pressure relaxation. Hypertension is the major cause of cardiac hypertrophy and heart failure. The pathological mechanism of cardiac pathological changes by hypertension has high connection with mitochondrion dysfunction and antioxidant ability of cardiomyocytes. The major of this study is to explore and discuss the efficacy and possible mechanisms of Morus alba .extract in preventing hypertensive cardiomyopathy. This study used 14 weeks old Spontaneously hypertensive rate (SHR) and Wistar-Kyoto rat (WKY) as models, Animals were fed with different concentration of Mori Folium extract for 8 weeks. Cardiac pathology and antioxidant defense system including the concentration of GSH and GSSG, activity of CAT, GPx, GR and SOD, and to analysis their gene expression of hypertrophy markers: ANF, α-MHC and β-MHC mitochondrial biogenesis factors: PGC-1, mtTFA and Pol-γ by RT-PCR were determined. The results showed that the CAT and GPX was lower and the ratio of GSH/GSSG was increased by Morus alba extract in WKY, and the expression of cardiac hypertrophy markers ANF was upregulated, but α-MHC, β-MHC mtTFA and PGC-1 were not affected by Morus alba extract. For SHR, Morus alba extract increased the activity of SOD and GR, and the ratio of GSH/GSSG, but no effect on the activity of CAT and GPx, and the gene expression of SOD and CAT was decreased. The high or middle dose of Morus alba extract enhanced the gene expression of SOD and CAT, but inhibition was fund in low dose, and the gene expression of Pol-γ decreased by increasing the dosage of Morus alba extract. In conclusion, the effect of antioxidant enzyme in SHR is stronger than WKY, and it also helps to regulate mitochondrial biogenesis of SHR. The Morus alba L. extract may increase antioxidant enzyme and decrease the oxidative stress of heart, and has the potential to prevent the hypertensive cardiomyopathy. |
