Popis: | 96 Owing to lacking the distribution of vessels in the cartilage and few chondrocyte in human it is more difficult to repair cartilage than other tissue for living organisms. Presently, allograft transplantation is a type of common surgery to repair cartilage. However, the amount of chondrocyte would depend on the three-dimension scaffold which would offer favorable conditions for chondrocyte growth in vitro. Therefore, several researchers investigated the creation of three-dimension scaffold to culture the chondrocyte. The micro-particles constituting three-dimension scaffold were successfully synthesized in vitro by simulating the beneficial and natural conditions for hyaline cartilage growth. During the synthesis of micro-particles, the stability of micro-particles was increased by adding the 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide cross linking agents for decreasing the degrading rate in vivo. In this study, the collagen and gelatin extracted from Taiwan Tilapia were substituted for the major components of human hyaline cartilage. Additionally, the poly micro-particle scaffold would be constructed as a result of the collagen and gelatin interacting with hyaluronic acid and chondroitin-6-sulfate. Finally, the poly micro-particle scaffold would be grinded into the micro-particles and micro-particles would be analyzed by differential scanning calorimetry, IR, and scanning electron microscope. |
