Study of Protocol on the Cryopreservation of Four Taiwan Medicinal Herbs - Lycopus lucidus Turcz., Scutellaria taiwanensis C.Y.Wu, Mosla chinensis Maxim., Blumea lacera (Burm.f.) DC. by Vitrification.

This research uses four Taiwanese medical herbs Lycopus lucidus Turcz., Scutellaria taiwanensis., Mosla chinensis Maxim., and Blumea hieraciifolia as materials to investigate the effects of survival rates of cryopreservation treated by vitrification. Plantlets of Lycopus lucidus Turcz. that have been cultured on MS medium containing 0.3 M sucrose for 30 days were treated with a loading solution (LS) for 60 minutes and dehydrated with a plant vitrification solution (PVS2) for 180 minutes. The results showed the best survival rates of 95.72%. Plantlets of Scutellaria taiwanensis which have been cultured on 1/2 MS medium containing 0.2 M sucrose for 3 days were treated with LS for 30 minutes and dehydrated with PVS2 for 80 minutes. The best survival rates of 50% was obtained. Plantlets of Mosla chinensis Maxim. which have been cultured on 1/2 MS medium containing 0.2 M sucrose for 7 days were treated with LS for 30 minutes and then dehydrated with PVS2 for 40 minutes. The best survival rates was 71.52%. Plantlets of Blumea hieraciifolia which have been cultured on MS medium containing 0.3 M sucrose for 20 days were treated with LS for 60 minutes and then dehydrated with PVS2 for 80 minutes, and the best survival rates was 70%. Physiological analysis of Lycopus lucidus Turcz. showed reduced relative water content, water potential and osmotic potential reduce and increased soluble sugar content, sucrose content and soluble protein content. These results indicate that Lycopus lucidus Turcz. culturing with high concentrations of sucrose results in dehydration of shoot tips and accumulation of solutes. The preculture procedures render increasing survival rates after cryopreservation due to the acclimatization in osmotic stress and increase freezing-tolerance for cryopreservation. This experiment suggests the protocol for four Taiwanese medical herbs cryopreservation by vitrification and can be served as a reference for long-term conservation of rare plant species.
