Allergic Effects of Six Pesticide Vehicles in Skin of Mouse

Allergic contact dermatitis (ACD) is a kind of atopic dermatitis that skin inflammation was induced by allergen. The allergen produce can directly harms the skin tissue, and form specific allergic symptoms. Most of the animal testing models for allergy require a log experimental time for about 3-4 months. It is not suitable to evaluate the potentical of pesticide vehicles to cause dermal sensitizing effects. Therefore, our experiments were carried out according to the internationally approved mouse ear swelling test (MEST) and lcoal lymph node assay (LLNA) motheds, using mouse to evaluate the dermal sensitization of pesticides inert ingredients, including n-methylpyrrolidinone (NMP), triphenyl phosphate (TPP), cyclohexanone (Cyc), diethylene glycol monomethyl ether (DGME), diethylene glycol monoethyl ether (DGEE) and diethylene glycol monobutyl ether (DGBE). The well known dermal sensitizing chemical, oxazolone (OXZ), was used as the positive control. Balb/c mice were given topical application of test chemicals to abdominal skin 3 times during induction phase and received challenging application to ears on Day 10 as challenge phase. Hypersensitivity reactions were evaluated by measuring ear swelling of MEST and lymphocyte proliferation of LLNA. Experiments also analyzed the pathological changes, the cytokines and chemokines change in ear skin. The results showed that OXZ has greater than 46% increase in thickness of the left ear than the right ear at 48th hour after challenge phase. Ear thickness of NMP group also increased greater than 31%. The histopathological observations revealed abumdant infiltration of eosinophils and mast cells in the dermisof left ear skin of OXZ and NMP. The cytokines (TNF-α and IL-6) and the chemokines (MIP1-α and Eotaxin) tests also showed significant increases in IL-6 and Eotaxin in OXZ treated group, and MIP1-α in NMP treated group. In LLNA test, results showed that stimulation index (SI) value was greater than 3, which occurred only in OXZ treated group. As to other five pesticide vehicles, there were no significant responses observed in LLNA test. The comprehensive tests carried out in this study showed successful the MEST and LLNA by using OXZ, as a postive contral. These models were applied to be rapidly screen of the large amount of pesticide vehicles. These data demonstrated that compared to six other ingredients in pesticide products, NMP might be more cutaneous allergericity to the Balb/c mice and could be potential risks to the human.
