Carbamazepine(CBZ), similar to tricyclic antidepressants, is an anticonvulsant used to treat partial seizures and tonic-clonic seizures. However, it is a poor water-soluble drug. Literatures indicate that the CBZ exhibits four different crystal forms due to different manufacturing way. In this study, we used DSC (Differential scanning calrimetry) to identify commercial CBZ’s crystal form first. Then we analyzed the identified crystal form of CBZ dissolved in different pH of 0.005M PBS buffer. The solubility of the CBZ in different concentration of taurocholate(TC)、α-cyclodextrin(α-CD)、β-cyclodextrin(β-CD)、sulfobutyl etherβ-cyclodextrin(SBE-β-CD) and hydroxypropyl β-cyclodextrin(HP-β-CD) was also investigated. Results indicate that solubility of the CBZ has no obvious change in different pH conditions including TC and α-CD. In contrast, addition of β-CD, especially its derivative HP-β-CD greatly improves CBZ’s solubility. Also the initial dissolution rate of a solution containing HP-β-CD is faster than that of other solutions. The information can help to formulate CBZ in pharmaceutics and increase its bioavailability.
