Gene regulation of the human oxytocin gene proximal promoter by the TR4 nuclear receptor

Popis: | 94 The human testicular receptor 4 (TR4) is a member of the nuclear receptor superfamily that shows a broad tissue distribution with higher expression in the nervous system. TR4 directly modulates gene expression through binding with the hormone response element (HRE) with the AGGTCA direct-repeat (DR). Some members of the nuclear receptor superfamily are able to regulate the transcriptional activity of the human oxytocin (OXT) gene promoter by binding to the multiple hormone response element, that has the AGGTCA DR sequence at approximately -160 upstream from the transcription start site of the OXT gene. Here, we want to study whether the TR4 can bind to the DR0 or double DR4 (dDR4) sequence on the OXT-HRE and further to study its influence. First, we used electrophoretic mobility shift assay to identify that TR4 could bind to the HRE on the OXT proximal promoter, and that also bind to the DR0 and dDR4 of the HRE. When we used unlabeled OXT-HRE, TR4 were titrated from the labeled OXT-HRE. We also used the luciferase reporter gene assay to study the interaction of TR4 on OXT via HRE. The results demonstrated that TR4 could activate OXT through both DR0 and dDR4 of the OXT-HRE. Thus, according to the results above, we proved that TR4 can regulate the expression of oxytocin gene through the binding to OXT-HRE. |
